加味麻杏石甘汤对EMT过程中ERK/GSK3β/snail信号通路的调控作用探讨

         

摘要

Objective To?explore?modified?maxing?shigan?decoction?played?a?role?in?the?ERK/GSK3β/snail?signaling?pathway?regulating?the?EMT?process.?Methods 18?cleaning?grade?SD?rats?were?randomly?divided?into?2?groups,12?in?Chinese?medicine?group?and?6?in?normal?control?group.?The?serum?of?Chinese?herbal?compound?was?prepared?according?to?15?times?of?the?clinical?adult?dose.?The?rats?were?given?two?doses?of?medicine?each?day,and?the?drug?was?given?three?times?in?a?row.?Cell?culture,ray?irradiation,frequency?of?3.64Gy/min,the?total?dose?was?5Gy.?The?cell?phenotype?protein?and?signal?pathway?main?protein?were?tested?by?Western?blot.?The?laser?confocal?microscopy?was?observed?in?the?snail.?Results? Compared?with?the?model+normal?rat?serum+ERK?inhibitors,model+the?drug-containing?serum/ERK?inhibitors,p-ERK,GSK-3βl,q?concentrations?were?significantly?lower,E-cadherin?concentration?increases,the?difference?was?statistically?significant(P<0.05).?Conclusion Modified?maxing?shigan?decoction?can?reduce?the?occurrence?of?EMT?by?regulating?the?expression?of?snail.?The?addition?of?ginkgo?may?control?the?gsk-3?beta?/snail?pathway?through?ERK?and?other?channels?to?inhibit?the?EMT?process.%目的 探讨加味麻杏石甘汤干预ERK/GSK3β/snail信号通路对上皮间质转化(EMT)过程的调控作用.方法 清洁级SD大鼠18只,随机分为2组,中药组12只,正常对照组6只,根据临床成人剂量的15倍制备中药复方含药血清,大鼠给药2次/d,连续给药3d,末次给药后2h采血;细胞培养,射线照射,频率为3.64Gy/min,总剂量为5Gy;Western?blot检测细胞表型蛋白及信号通路主要蛋白(E-cadherin、Vimentin、α-SMA、Snail);激光共聚焦显微镜观测snail转核.结果 与模型+正常大鼠血清+ERK抑制剂组比较,模型+含药血清+ERK抑制剂组,p-ERK、GSK-3β、snail浓度均明显下降,E-cadherin浓度升高,差异有统计学意义(P<0.05).结论 ERK抑制剂能够显著抑制靶基因的表达,减弱EMT过程.加味麻杏石甘汤可能通过ERK及其他途径调控GSK-3β/snail通路,抑制EMT过程.
