人参二苓汤及其水提醇沉提取物对H22荷瘤小鼠脾胸腺指数及外周血细胞的影响

         

摘要

目的:探讨人参二苓汤及其水提醇沉提取物对H22荷瘤小鼠脾胸腺指数及外周血细胞的影响。方法将60只健康昆明小鼠分为正常组、荷瘤模型组、阳性对照组(CTX组)、水提组(RSEL)、总多糖组(PRSEL)、小分子组(SRSEL),每组10只。建立H22荷瘤小鼠模型,造模成功后分别灌服人参二苓汤水提物、总多糖、小分子,实验结束后计算小鼠脾胸腺指数,抑瘤率及外周血细胞的检测。结果人参二苓汤水提物(RSEL)组和总多糖(PRSEL)组抑瘤率分别为37.80%和50.13%;CTX组为79.62%;CTX组及人参二苓汤水提组和总多糖组平均瘤重分别为(0.7611±0.1502)g、(2.3231±0.5324)g和(1.8642±0.5740)g,与荷瘤模型组平均瘤重(3.7352±0.4851)g比较,差异有统计学意义(P<0.01),小分子组瘤重与荷瘤组比较差异无统计学意义(P>0.05);与水提组比较,总多糖组瘤重差异有统计学意义(P<0.05);与模型组比较,人参二苓汤总多糖(PRSEL)组能显著增强荷瘤小鼠脾和胸腺指数(P<0.05或P<0.01),且较人参二苓汤水提物(RSEL)组和小分子(SRSEL)组更为显著;人参二苓汤总多糖(PRSEL)组能改善荷瘤小鼠晚期白细胞的过度增高状态(P<0.01),优于人参二苓汤水提物组(P<0.05)。结论人参二苓汤总多糖(PRSEL)可以明显延缓荷瘤小鼠晚期类白血病样反应的发生,维持血细胞在一个相对稳定的状态,且抑制肿瘤生长作用较明显,可能与其改善荷瘤小鼠免疫器官功能相关。%Objective To investigate the Renshen Erling decoction and its water extract from alcohol precipita-tion on spleen index and thymus index and peripheral blood cells in H22 tumor-mice. Methods Sixty healthy KM mice were randomly divided into 6 groups: control group, tumor-bearing model group, positive control group (CTX group), Renshen Erling decoction-water extract group (RSEL), Renshen Erling decoction-total polysaccharide group (PRSEL), and Renshen Erling decoction-small molecule group (SRSEL), with 10 mice in each group. H22 tumor-bearing model was established in all group except control group, then mice in each group were treated with the corresponding drugs for 14d. On day 15, mice in each group were sacrificed for calculation of spleen and thy-mus index and inhibitory rate and counting of peripheral blood cells. Results The inhibitory rate were 37.80%, 50.13%, and 79.62% in RSEL group, PRSEL group and CTX group, respectively; the average tumor weight in CTX group, RSEL group, and PRSEL group were 0.7611 ±0.1502g, 2.3231 ±0.5324g and 1.8642 ±0.5740g, respectively, with a significant difference compared to tumor-bearing model group (3.7352±0.4851g, P<0.01); no significant dif-ference was found between that of SRSEL group(3.5072±0.278g) and tumor-bearing model group(P>0.05); a sig-nificant difference was noted between RSEL group and PRSEL group(P<0.05). The spleen index and thymus index of PRSEL group were higher than that of tumor-bearing model group(P<0.05 or P<0.01) and even higher than that of RSEL and SRSEL groups. The excessive increase of leucocyte in advanced tumor-bearing mice was inhibited in PRSEL group(P<0.01) and the inhibition effect was better compared to that in RSEL group(P<0.05). Conclusion Renshen Erling decoction-PRSEL can significantly delay the leukemoid reaction of tumor-bearing mice and main-tain their blood cells in a relatively stable state. Its effect on inhibiting tumor growth is clear, which may be relat-ed to its role in improving the function of immune organs in mice.
