Sodium dodecyl sulfate-polyacrylamide gelnelectrophoresis (SDS-PAGE) is a universally employedntechnique that separates proteins on the basis of molecularnweight (MW). However, membrane proteins are known tonsize anomalously on SDS-PAGE calibrated with conven-ntional standards, an issue that complicates interpretation ofnprotein identity, purity, degradation, and/or stoichiometry.nHere we describe the preparation of novel polyleucinenhydrophobic standards for SDS-PAGE that reduce thenaverage deviation of the apparent MW from the formulanMWof natural membrane proteins to 7%versus 20%withncommercially available standards. Our results suggest thatngel calibration with hydrophobic standards may facilitate theninterpretation ofmembrane protein SDS-PAGE experiments.
