Chemical Composition, Antifungal and Radical Scavenging Activities of Egyptian Mandarin Petitgrain Essential Oil

The composition and biological activities of Egyptian mandarin petitgrain essential oil and its fractions are reported. The gas chromatographic-mass spectroscopic (GC/MS) study of this oil and its oxygenated and non-oxygenated fractions revealed that the major components were methyl-n-methyl anthranilate (62.82%), m-terpincne (17.2%), p-cymene (3.52%) and α-pinene (2.67%) for total oil; methyl-n-methyl anthranilate (85.89%), caryophyllene oxide (1.85%) and hexdecaoic acid (1.37%) for oxygenated fraction compared to γ-terpinene (52.65%). caryophellene (10.3%), α-pinene (6.1%), p-cymene (5.91%) and gurunene (4.47%) for non-oxygenated fraction. The antifungal efficacy of the mandarin oil and its fractions when examined by disc diffuxion test at different comcentrations showed that the mandarine oil and its oxygenated fraction (at 75 μL) strongly inhibited the growth of all the tested fungi (75-99%). This activity might be due to the presence of high content of methyl-n-methyl anthranilate. In contrast, the non-oxygenated fraction was less effective against the tested fungi (5-26% inhibition) under the same conditions. The mandarin petitgrain oil and its fractions were further investigated for radical scavenging activities using 1,1-diphenylpicryl-2-picrylhydrazyl (DPPH). At a concentration of 2500 ppm, all the tested samples showed radical scavenging activity. Obviously, the oxygenated fraction (mainly methyl-n-methyl anthranilate) exhibited higher significant reduction of DPPH radicals (86%) as compared with BHA (85%) and gallic acid (76%). The present results support the view that the Egyptian mandarin petitgrain essential oil is highly bioavailable and may be considered as safe flavouring agent with pleasent aroma in addition to being antioxidant with free radical scavengers. Also its biological activity is shown to be particularly interseting for food and cosmetics industries.
