The human e-globin gene is expressed in a tissue-specific and developmental stage-specific manner. During the earliest stage of gestation, this gene is expressed in the yolk sac, but is silenced completely at the 6th—8th weeks of gestation. Recently,several studies on the transgenic mice have shown that the 5'-fianking DNA sequences of human e-globin gene are very important in the tissue- and stage-specific regulation of human e-globin gene expression. Both the positive regulatory element (from —535 to — 453 bp) and the silencer (from —392 to — 177bp) upstream of human e-globin gene have been identified (fig. 1). However, the molecular mechanisms involved in both the activation and repression of human e-globin gene expression are unclear.
