Fluorescence Correlation Spectroscopy (FCS) in itsrnoriginal form was applied first to measure the diffusion and chemical kinetics of DNA-drug intercalation, wherein it was used as a special case of relaxation analysis. In the following years it was extender to different areas such as determining particle concentration, study of translational and rotational mobility in two or three dimensions and similar areas. The early measurements suffered from poor signal-to-noisc ratios, mainly because of low detection efficiency, large ensemble numbers and insufficient background suppression. It has recently experienced growing popularity in biophysical and biochemical applications due to significantly improved signal-background ratios and single molecule sensitivity. It is one of the many different modes of high resolution spatial and temporal analysis of extremely low concentration biomolecules. Its versatility lies in the fact that all physical parameters giving rise to fluctuations in the fluorescence signal are accessible by the FCS.
