Metabolicsyndrome(MetS),acompilationofassociatedriskfactors,increasestheriskoftype2diabetesandcoronaryarterydisease(CAD,atherosclerosis),whichcanprogresstothepointofarteryocclusion.StentsaretheprimaryinterventionaltreatmentforocclusiveCAD,andpatientswithMetSandhyperinsulinemiahaveincreasedrestenosis.Becauseofitsthriftygenotype,theOssabawpigisamodelofMetS.Wetestedthehypothesisthat,whenfedhigh-fatdiet,OssabawswinedevelopmorefeaturesofMetS,greaternativeCAD,andgreaterstent-inducedCADthandoYucatanswine.Animalsofeachbreedweredividedrandomlyinto2groupsandfed2differentcalorie-matcheddietsfor40wk:controldiet(C)andhigh-fat,high-cholesterolatherogenicdiet(H).Abaremetalstentwasplacedinthecircumflexartery,andpigswereallowedtorecoverfor3wk.CharacteristicsofMetS,macrovascularandmicrovascularCAD,in-stentstenosis,andCasupxmlns="http://pub2web.metastore.ingenta.coms/"2+/supsignalingincoronarysmoothmusclecellswereevaluated.MetScharacteristicsincluding,obesity,glucoseintolerance,hyperinsulinemia,andelevatedarterialpressurewereelevatedinOssabawswinecomparedtoYucatanswine.OssabawswinewithMetShadmoreextensiveanddiffusenativeCADandin-stentstenosisandimpairedcoronarybloodflowregulationcomparedwithYucatan.In-stentatheroscleroticlesionsinOssabawcoronaryarterieswerelessfibrousandmorecellular.CoronarysmoothmusclecellsfromOssabawhadimpairedCasupxmlns="http://pub2web.metastore.ingenta.coms/"2+/supeffluxandintracellularsequestrationversuscellsfromYucatanswine.Therefore,OssabawswineareasuperiormodelofMetS,subsequentCAD,andcellularCasupxmlns="http://pub2web.metastore.ingenta.coms/"2+/supsignalingdefects,whereasYucatanswineareleanerandrelativelyresistanttoMetSandCAD.
