Meal-Feeding Studies in Mice: Effects of Diet on Blood Lipids and Energy Expenditure

Toidentifyoptimalstudy-designconditionstoinvestigatelipidmetabolism,male,C57BL/6Jmice(age,59±3days)wereallottedtoeightgroups,withsixanimalspergroupthatwerestratifiedbythreefactors:diettype(highfat[HF]:60%ofenergyfromfatversusthatofastandardrodentdiet,14%fat,fedfor7weeks),feedingregimen(adlibitum[adlib]versusmealfed),andmetabolicstate(datacollectedinfastedorfedstates).Serumfreefattyacids(FFA)andtriacylglycerols(TAG)concentrations,andenergyexpenditure(EE)wereassessed.Micegained0.30±0.11gofbodyweight/daywhenallowedadlibaccesstoHFdiet,similarweightwhenmeal-fedtheHForadlib-fedthestandarddiet(0.10±0.03g/day),andnoweightwhenmeal-fedthestandarddiet(0.01±0.02g/day).Fed-stateTAGconcentrationwas88to100%higher(P0.02)thanthatofthefastedstate,exceptwhenanimalswereadlib-fedtheHFdiet.Whenthestandarddietwasmealfed,FFAconcentrationwas30%higherinthefastedcomparedwiththefedstate(P=0.003).Micehad33%higherpostprandialEEwheneitherdietwasmealfed(P=0.01).Miceadaptedtomealfeedingdevelopedtransitionsinmetabolismconsistentwithknownphysiologicchangesthatoccurfromfastingtofeeding.Whenfedthestandarddiet,a6-hperdaymeal-feedingregimenwasrestrictivefornormalgrowth.Thesedatasupportuseofameal-feedingregimenwhenHFdietsareusedandresearchisfocusedonmetabolicdifferencesbetweenfastedandfedstates.ThisprotocolallowsstudyofthemetaboliceffectsofanHFdietwithouttheconfoundingeffectsofover-consumptionoffoodandexcessbodyweightgain.
