Ulcerativedermatitis(UD)isacommoncauseofmorbidityandeuthanasiainmicewithaC57BL/6(B6)background.ThepurposesofthecurrentstudyweretodeterminewhetherUDlesionscouldbereliablyproducedinB6micelackingstearoyl-CoAdesaturase1(SCD1sup#8211;/#8211;/supmice),toascertainwhethertheUDlesionsinSCD1sup#8211;/#8211;/supmiceweresimilartothosefoundinotherB6mice,andtocharacterizethecellinvasionphenotypeofStaphlococcusxylosusculturedfromthelesions.S.xylosusisolatesfromtheenvironmentandhumanskinwereusedascontrols.SCD1sup#8211;/#8211;/sup(n=8pergroup)andnontransgenicB6controlmice(n=22micepooledfrom3groupsthatreceiveddifferentconcentrationsofconjugatedlinoleicacid)werefedstandardrodentchoworasemipurifieddiet(NIHAIN76A)for4wk.SamplesfromotherB6micewithUD(fieldcases;n=7)alsoweresubmittedforhistologyandculture.AlloftheSCD1sup#8211;/#8211;/supmicedevelopedUDlesionsby4wkonNIHAIN76A.NoneofSCD1sup#8211;/#8211;/supfedstandardrodentchowandnoneofthewildtypeB6micefedNIHAIN76AdevelopedUD.Supplementationwithconjugatedlinoleicaciddidnotaffectulcerogenesis.UDlesionsinSCD1sup#8211;/#8211;/supmiceandfieldcasesweregrosslyandhistologicallysimilar.S.xylosuswasisolatedfromSCD1sup#8211;/#8211;/supmicewithUD(71%)andfieldcasesofUD(43%).Theseisolateswerethemostcell-invasive,followedbytheenvironmentalisolate,andfinallythehumanskinisolate.OurresultsprovideabasisforfurtherpathologicandclinicalstudyofUD.
