Thisstudyinvestigatedwhetherdietarysupplementationofpolyphenolics-richgrapeextract(GE)couldattenuateendotoxin-inducedserumsecretoryphospholipaseAsub2/sub(sPLAsub2/sub)activity,amodulatorofinflammation.MaleSprague#8211;Dawleyratswerefedacontroldietorthedietsupplementedwithpolyphenolic-richGE(100or300mg/kgdaily)for3wkpriortointraperitonealinjectionof3or15mg/kgLPS.AfluorometricassaywasusedtomeasureserumsPLAsub2/subactivityduringa5-dperiodbeforeandafterLPSinjection.Bodyweight,hematocrit,andserumC-reactiveproteinlevelwerealsomeasured.AdministrationofLPSinducedarapidincreaseinsPLAsub2/subactivity,whichpeaked1to2dafterLPSinjectionandresolvedtonear-baselinevaluesondays4to5.Markeddeclinesinbodyweightandhematocrit,increasesinC-reactiveproteinlevels,andeffectsonhealthstatusalsooccurred.GEsupplementationsignificantlyattenuatedtheLPS-inducedincreaseinsPLAsub2/subactivityanddeclineinhematocrit,butitseffectsonthelossofbodyweightandC-reactiveproteinlevelswerenotsignificant.Amongthemeasurements,serumsPLAsub2/subwastheonlymarkerthatshowedadose-dependentresponsetobothLPSandGEsupplementation.Thecurrentfindingsshowthatoralconsumptionofpolyphenolic-richGEsuppressesendotoxin-inducedsPLAsub2/subactivity.
