Maspin(serpinB5),atumor-suppressingmemberoftheserineproteaseinhibitorfamily,participatesincellmigration,adhesion,invasion,andapoptosis.Theseprocessesarealsocriticalforembryoimplantation,buttheroleofmaspininembryoimplantationremainspoorlyunderstood.Theaimofthepresentstudywastoinvestigatethespatiotemporalexpressionofmaspininearlypregnantmouseendometriumanditsroleinembryoimplantation.Real-timequantitativePCRanalysis,immunohistochemistry,andWesternblottingwereusedtodetectmRNAandproteinexpressionofmaspinintheendometriaofnonpregnantandearlypregnant(days0to7)mice.Onday3ofpregnancy,miceinthetreatedgroup(n=20)wereinjectedintheleftuterinehornwithantimaspinpolyclonalantibodyandintherighthornwithpurifiedrabbitIgG;controlmice(n=20)wereinjectedonlywithpurifiedrabbitIgGintherightuterinehorn.Implantedembryoswerecountedonpregnantday8.ThemRNAandproteinexpressionsofmaspinwerehigherintheendometriaofpregnantmicethannonpregnantmice;theselevelsgraduallyincreasedfromday1ofpregnancy,peakedonday5,andthendecreasedondays6and7.Themicetreatedwithantimaspinpolyclonalantibodygrouphadfarfewerimplantedembryosthandidthecontrolgroup.Takentogether,theseresultssuggestthatmaspin,atumorsuppressor,mayplayanimportantroleinembryoimplantation.
