Mutationalactivationofthegeneforepidermalgrowthfactorreceptor(EGFR)is1ofthemainwaysbywhichthisreceptorinducesnon-smallcelllungcancers(NSCLC).VariantIIIEGFR(EGFRvIII)isapotentialtherapeutictargetinNSCLCtreatmentbecauseofthehighfrequencyofdeletionmutationsinthisprotein.Thisstudyusednoninvasivemagneticresonanceimaging(MRI)toinvestigatetheroleofanEGFRvIIImutantinlungtumorigenesisandtumormaintenanceaswellasitsresponsetotheEGFRsmallmoleculeinhibitorerlotinib(Tarceva)onbitransgenicmice.Bothspin-echoandgradient-echosequenceswithandwithoutcardiacandrespiratorygatingwereperformedtoimagetheinvasivemouselungtumordrivenbyEGFRvIIImutation.Tumorvolumesweremeasuredbasedon2-dimensionalaxialMRI;3-dimensionalrenderingoftheimageswereobtainedtodemonstratethespatiallocationanddistributionofthetumorinthelung.TheMRIresultsindicatedthatthetumordrivenbytheEGFRvIIImutationwasgeneratedandmaintainedinthebitransgenicmicewiththeuseofdoxycycline.TumormonitoringviaMRIshowedthatErlotinibcansignificantlyinhibitthegrowthoftumorinvivo.MRIhastheabilitytoimagemouselungtumorwithdifferentsequencesfocusingontissuecontrastsbetweentumorandsurroundings.TheMRIapproachesinthisworkcanbeappliedonotherantitumordrugtreatmentevaluationinvivowhenappropriatesequencesarechosen.
