TheNIHguidelinesforsurvivalbleedingofmiceandratsnotethatusingtheretroorbitalplexushasagreaterpotentialforcomplicationsthandoothermethodsofbloodcollectionandthatthisprocedureshouldbeperformedonanesthetizedanimals.Lateralsaphenousveinpuncturehasalowpotentialforcomplicationsandcanbeperformedwithoutanesthesia.Mongoliangerbils(Merionesunguiculatus)arethepreferredrodentmodelforfilarialparasiteresearch.Tomonitormicrofilariacountsintheblood,bloodsamplingfromtheorbitalplexushasbeenthestandard.Ourgoalwastorefinethebloodcollectiontechnique.Todeterminewhetherbloodcollectionfromthelateralsaphenousveinwasafeasiblealternativetoretroorbitalsampling,wecomparedmicrofilariacountsinbloodsamplescollectedbybothmethodsfrom21gerbilsinfectedwiththefilarialparasiticwormBrugiapahangi.Lateralsaphenousveincountswereequivalenttoretroorbitalcountsatrelativelyhighcounts(greaterthan50microfilariaeper20#956;L)butweresignificantlylowerthanretroorbitalcountswhenmicrofilarialconcentrationswerelower.Ourresultsindicatethatalthoughretroorbitalcollectionmaybepreferablewhenlowconcentrationsofmicrofilariaeneedtobeenumerated,thelateralsaphenousveinisasuitablealternativesiteforbloodsamplingtodeterminemicrofilaremiaandisafeasiblerefinementthatcanbenefitthewellbeingofgerbils.
