Theoriginoftheage-associateddegenerativeprocessesinmeniscaltissueispoorlyunderstoodandmayberelatedtoanimbalanceofanabolicandcatabolicmetabolism.Theaimofthecurrentstudywastocomparemedialmenisciisolatedfromjuvenilepigsanddegeneratedmedialmeniscifromadultpigsintermsofgeneexpressionprofileandultrastructure.Medialmenisciwereisolatedfromthekneejointsofjuvenileandadultpigs(n=8foreachgroup).Degenerationwasdeterminedhistologicallyaccordingtoascoringsystem.Inaddition,thegeneexpressionprofilesof14genesencodingextracellularmatrixproteins,catabolicmatrixmetalloproteinasesandmediatorsofinflammationwereanalyzed.Changesintheultrastructureofthecollagennetworkofthemeniscaltissuewereanalyzedbyusingtransmissionelectronmicroscopy.Thehistologicanalysisofmeniscishowedsignificantlyhighergradeofdegenerationintissueisolatedfromadultporcinekneejointscomparedwithmenisciisolatedfromjuvenilekneejoints.Inparticular,destructionofthecollagennetworkwasgreaterinadultmeniscithaninjuvenilemenisci.DegeneratedmeniscishowedsignificantlydecreasedgeneexpressionofCOL1A1andincreasedexpressionofMMP2,MMP13,andIL8.Themeniscifromadultporcinekneejointscanserveasamodelformeniscaldegeneration.Degenerativechangesweremanifestedasdifferencesinhistopathology,geneexpressionandultrastructureofcollagennetwork.
