Thestudyaimedtorevealalterationsinexpressionandmethylationlevelsofthegrowth-relatedimprintedgenesH19andIgf2infetusesofdiabeticmice.Diabeteswasinducedinfemalemicebyintraperitonealinjectionofstreptozotocin.DNAandtotalRNAwereextractedfromfetusesobtainedfromdiabeticandcontroldamsonembryonicday(E)14.Real-timeRT-PCRanalysisrevealedthatthemRNAexpressionofIgf2infetusesfromdiabeticmicewas0.65-foldofthecontrolcounterparts.BisulfitegenomicsequencingdemonstratedthatthemethylationleveloftheH19#8211;Igf2imprintcontrolregionwas19.1%higherindiabeticfetusesthaninthoseofcontroldams.Inaddition,thebodyweightofpupsborntodiabeticdamswas26.5%lowerthanthatofthecontrolgroup.Theresultsindicatethatmaternaldiabetescanaffectfetaldevelopmentbymeansofalteredexpressionofimprintedgenes.ThemodifiedgenomicDNAmethylationstatusofimprintinggenesmayaccountforthechangeingeneexpression.
