Issuesofcostandgeneticscanresultininbreedingofcaninegeneticdiseasecolonies.Beaglesoftenareusedtomaintainsuchcolonies,providingstockforoutcrosses.FactorVII(FVII)deficiencyisahemostaticdisorderfoundatincreasedfrequencyinbeaglesandhasbeencharacterizedattheDNAlevel.DeficiencyofFVIIpresentsobstaclesincoloniesfoundedwithbeagles.AninitialfindingofaFVII-deficientpupfromalongstandingcolonypromptedustoevaluateFVIIdeficiencyfullyinthiscolony.Currentandarchivalrecordsandtissueswereusedtoreconstructthecolonypedigree,assessthecontributionfrombeagles,andtestsamplestodocumentthesourceandfrequencyofthemutantFVIIallele.AspartofthisstudywedevelopedaPCR-baseddiagnosticassaythatwassimplerthanwhatwaspreviouslyavailable.Pedigreeanalysisrevealedafoundereffectimplicatingbeaglesthatledtohighfrequency(55%)ofthemutantallele.Inaddition,affectedanimalswereidentified.Thecompletepictureoftheclinicaleffectwithinthecolonyremainsunclear,butunusualneonatalpresentations,includinghemoabdomen,haveoccurredinpupsaffectedwithFVIIdeficiency.UseofaPCR-baseddiagnosticassaytoscreenallpotentialbeaglebreedingstockwillpreventsimilaroccurrencesofFVIIdeficiencyinfuturecanineresearchcolonies.
