Hemotrophicmycoplasma(hemoplasma)infectioninresearchsheepcanconfoundexperimentalresultsandcontributetomorbidityandmortality.Prevalenceandclinicopathologicstudieshistoricallyreliedonblood-smeardiagnosis,butsystematicstudiesusingcurrentmoleculartechniquesarewarranted.Herewesoughttoreporttheprevalenceofsubclinicalinfectioninourstudypopulation,comparediagnosticsensitivityandspecificitybetweenbloodsmearsandaPCRassay,anddeterminetheeffectsofinfectiononCBCvariablesanderythrocytemembranefragility.Wecollectedwhole-bloodsamplesfrom111convenience-sampledresearchsheep.AllsamplesweretestedforhemoplasmasbyusingaPCRassay,bloodsmearswereevaluatedforvisualpresenceofhemoplasmas,andCBCandosmoticfragilityassayswereperformed.Subclinicalprevalence,accordingtoPCRdiagnosis,was14.1%(14of99)inourstudypopulation.RelativetothePCRassay,blood-smeardiagnosiswas8.3%sensitiveand100%specificforhemoplasmadetection.SubclinicalinfectionwasassociatedwithchangesinMCV,MCHC,RBCdistributionwidth,andabsolutemonocytecount.AcuteinfectionwasassociatedwithchangesinRBCmass,Hgbconcentration,MCV,MCH,MCHC,andabsolutelymphocyteandmonocytecounts.Acuteinfectionwasassociatedwithincreasedmeanerythrocytefragilitycomparedwiththatinuninfectedcontrolandtreatedsheep.Wedemonstratedthathemoplasmainfectioniscommoninourstudypopulation,blood-smearevaluationisinsensitiveatdetectinginfection,andinfectionisassociatedwithchangesinCBCvariablesandincreasederythrocytemembranefragility.Thesefindingsraiseconcernsregardingthesuitabilityofhemoplasma-infectedsheepforbiomedicalresearch.
