Mycoplasmosisisafrequentcausativemicrobialagentofcommunity-acquiredpneumoniaandhasbeenlinkedtoexacerbationofchronicobstructivepulmonarydisease.ThemacrophageclassAscavengerreceptor(SRA)facilitatestheclearanceofnoxiousparticles,oxidants,andinfectiousorganismsbyalveolarmacrophages.WeexaminedwildtypeandSRAsup#8211;/#8211;/supmice,housedineitherindividuallyventilatedorstaticfilter-topcagesthatwerecycledwithfreshbeddingevery14d,asamodelofgene#8211;environmentinteractionontheoutcomeofpulmonaryMycoplasmapulmonisinfection.IntracageNHsub3/subgasmeasurementswererecordeddailypriortoinfection.Micewereintranasallyinfectedwith1×10sup7/supcfuM.pulmonisUABCTandevaluatedat3,7,and14dafterinoculation.Wildtypemicecleared99.5%ofpulmonaryM.pulmonisby3dafterinfectionbutremainedchronicallyinfectedthroughthestudy.SRAsup#8211;/#8211;/supmicewerechronicallyinfectedwith40-foldhighermycoplasmanumbersthanwerewildtypemice.M.pulmoniscausedachronicmixedinflammatoryresponsethatwasaccompaniedwithhighlevelsofIL1#946;,KC,MCP1,andTNF#945;inSRAsup#8211;/#8211;/supmice,whereaspulmonaryinflammationinWTmicewasrepresentedbyamonocytosiswithelevationofIL1#946;.HousinghadaprominentinfluenceontheseverityandpersistenceofmycoplasmosisinSRAsup#8211;/#8211;/supmice.SRAsup-/-/supmicehousedinstaticcageshadanimprovedrecoveryandsignificantchangesinsurfactantproteinsSPAandSPDcomparedwithbaselinelevels.TheseresultsindicatethatSRAisrequiredtopreventchronicmycoplasmainfectionofthelung.Furthermore,environmentalconditionsmayexacerbatechronicinflammationinM.pulmonis-infectedSRAsup#8211;/#8211;/supmice.
