AsubstrainofmiceoriginatingfromtheCF1strain(anoutbredcolony)rearedatOsakaPrefectureUniversity(CF1/bcacmice)developscataractsbeginningat14dold.Affectedmicewerefullyviableandfertileandhaddevelopedcataractsby22dofage.Theincidenceofcataractsdidnotdifferbetweenmaleandfemalemice.Histologically,14-wk-oldCF1/bcacmiceshowedvacuolatedlensepithelialcells,swollenlensfibers,manypyknoticnuclei,andvacuolationofthelenscortex.Toelucidatethemodeofinheritance,weanalyzedheterozygousmutantshybridsgeneratedfromCF1/bcacandwildtypeBALB/cmiceandtheoffspringofthebackcrossedheterozygousmutants.Noneoftheheterozygousmutantswasaffected,buttheratioofaffectedtounaffectedmicewas1:3amongtheoffspringoftheheterozygousmutants.Theinitialgenomewidescreenof20affectedbackcrossedoffspring(CF1/bcac×[CF1/bcac×BALB/c])indicatedthatthemutantgeneresidesonchromosome16.Forfurthermapping,weusedaffectedprogenyofCF1/bcac×(CF1/bcac×MSM/Ms)mice.WeconcludedthatthecataractsinCF1/bcacmiceareinheritedthroughanautosomalrecessivemutationandthatthemutantgeneislocatedonmousechromosome16betweenD16Mit5andD16Mit92andbetweenD16Mit92andD16Mit201.ThemappingofthemutantgeneoftheCF1/bcacmicetomousechromosome16providesthepositionalinformationnecessarytoidentifythecandidategeneresponsiblefortheCF1/bcacphenotype.
