Geneticallyalteredmiceareimportantresearchtoolsforthestudyofhumandevelopmentanddisease.Occasionally,whetherornotrelatedtothegeneticmutation,micemaybecomeinfertilewithageand,thus,risklossofthemutantline.Underconditionsinwhichassistedreproductiontechniques(ARTs),suchasinvitrofertilization,areunsuccessful,anewstrategy,intracytoplasmicsperminjection(ICSI),maybeapplicable.Thistechniquehasbeenperfectedforuseinthemouseandisnowconsideredareliable,effective,andefficientART.Inthestudyreportedhere,we#8220;rescued#8221;(i.e.,producedoffspring,usingICSIfroma#8220;last-of-line#8221;mutantmalemouse)fourlinesthatotherwisehadbecomeinfertileandunresponsivetoconventionalART's.Atotalof26livepupswereproducedfromeightpregnantrecipientfostermothers.Fivemutantmalemicewerederived(oneeachfromthreelines,andtwofromoneline),andallsurvivedtoadulthood.WefoundthatlivebornmicecouldbesuccessfullyderivedbyuseofICSIthatsubsequentlycouldbreedbynaturalmatingtoreestablishthemutantline.Becauseofitseffectivenessandreliabilityundertheseconditions,ICSIshouldbeconsideredapowerfuladditiontothearmamentariumofART'sapplicableinthegenetically-alteredmouse,especiallywhenonlyonemalemaystillbeavailable.
