Ethanolisadietaryfactorthatdose-dependentlyincreasesbreastcancerriskinwomen.WepreviouslyhaveshownthatethanolincreasesmammaryepithelialdensitythroughincreasedbranchingafterdietaryexposureduringpubertyinCD2/F1mice.Toextendthesestudiestoparousmiceinabreastcancermodel,weusedatransgenicmousemodelofhumanparity-associatedbreastcancer,theFVB-MMTV-Her2/Neumouse,whichoverexpresseswildtypeEGFR2,resultinginconstitutiveactivationofgrowthsignalinginthemammaryepithelium.Herewedescribetheshort-termeffectsofethanolfeedingonprogressionthroughinvolution.Micewerefeddietssupplementedwith0%,0.5%,1%,or2%ethanolfor4,9,or14dstartingonday21oflactation(thatis,atthestartofnaturalpostlactationalinvolution).Unlikeperipubertalmiceexposedtoethanol,postlactationaldamsshowednochangesinbodyweight;liver,spleen,andkidneyweights;andpathology.Ethanolexposurehadnoeffectonmammaryglandlobulardensityandadipocytesizethroughoutinvolution.Likewise,theinfiltrationofinflammatorycellsandserumoxidizedlipidspecieswereunchangedbydiet,suggestingthatethanolfeedinghadnoeffectonlocalinflammation(leukocyteinfiltration)orsystemicinflammation(oxidizedlipids).Inconclusion,ethanolexposureofparousdamshadnoeffectonmammaryglandstructureortheregressionofthelactatingmammaryglandtoarestingstate.Theperiodofinvolutionthatfollowsnaturallactationappearstoberefractorytodevelopmentaleffectsofethanolonmammaryepithelium.
