ThenucleotidesubstitutionC797TintheChrm2genecausessubstitutionofleucineforprolineatposition266(P266L)oftheCHRM2protein.BecauseChrm2codesforthetype2muscarinicreceptor,thismutationcouldinfluencephysiologicandbehavioralphenotypesofmice.Chrm2mRNAwasnotdifferentiallyexpressedin2brainregionswithhighcholinergicinnervationinamousestrainthatdoes(BALB/cByJ)ordoesnot(C57BL/6J)havethemutation.Inaddition,strainsofmicewithandwithouttheC797TpointmutationinChrm2didnotdiffersignificantlyinmuscarinicbindingproperties.Variationacrossstrainswasdetectedintermsofacousticstartle,prepulseinhibition,andthephysiologiceffectsofthemuscarinicagonistoxotremorine.However,interstraindifferencesinthesemeasuresdidnotcorrelatewiththepresenceofthemutation.AlthoughwewereunabletoassociateameasurablephenotypewiththeChrm2mutation,assessmentofthemutationonothergeneticbackgroundsorinthecontextofothertraitsmightrevealdifferentialeffects.Therefore,despiteournegativefindings,evaluationofcharacteristicsthatinvolvemuscarinicfunctionshouldbeundertakenwithcautionwhencomparingmicewithdifferentallelesoftheChrm2gene.
