BackgroundandPurpose:Staphylococcusaureusisanimportantcauseofintravascularcatheter-associatedbacteremia.Wedevelopedaratcentralvenouscatheter(CVC)-associatedinfectionmodeltostudypathogenesisandtreatment.brxmlns="http://pub2web.metastore.ingenta.coms/"/brMethods:Asilasticlumen-within-lumencatheterandrodent-restraintjacketweredesigned.Subcutaneouslytunneledcatheterswereinsertedinthejugularveinof20maleSpragueDawleyrats.Twelverats(group1)wereinoculatedwithS.aureusviatheCVC;threerats(group2)wereinoculatedwithS.aureusviathetailvein,fiverats(group3)servedasuninfectedcontrols;andthreerats(group4)wereinoculatedwithS.aureusviathetailveinbutdidnotundergoCVCinsertion.Fivetoeightdaysafterinoculation,animalswereeuthanized,CVCswereasepticallyremoved,andquantitativeculturewasdone.Quantitativeculturealsowasperformedonblood,heart,liver,lungs,andkidneys.brxmlns="http://pub2web.metastore.ingenta.coms/"/brResults:Infection,characterizedbybacteremiaandmetastaticdisease,wasobservedinallratsinoculatedviatheCVCwithasfewas100colony-formingunits(CFU)ofS.aureus.Ratsofgroup2werenotaslikelytodevelopCVC-associatedinfection,andnoneoftheanimalsofgroups3or4developedinfection.brxmlns="http://pub2web.metastore.ingenta.coms/"/brConclusions:ThismodelofCVC-associatedinfectionshouldprovesuitableforstudyingpathogenesisandtreatmentofthecondition.
