Bacterial Lipopolysaccharide Induces a Conduction Block in the Sciatic Nerves of Rats

AsingleinjectionofEscherichiacolilipopolysaccharide(LPS;intraperitoneally[i.p.]andintravenously[i.v.])reliablyinducesperipheralnervedisturbancesinthehindlimbsofinbredAustralianalbinoWistar(AaW)rats.Intheseriesofexperimentspresentedhere,weaimedtocharacterizethissyndromebyexaminingelectrophysiologic,immunologic,andimmunochemicalfeatures.TheLPS-inducedneurologicsequelaeinAaWratsweretransient,atleastpartlyreversiblebydrugtreatment,andwerenotassociatedwithanydetectableneuropathologicfindingsbylightmicroscopy.Neurologicsequelaewerepreventedbyadministrationofdexamethasoneandbypretreatmentwiththemacrophageinhibitorgadoliniumchloride,suggestingthattheywerecausedbyLPS-inducedactivationofperipheralmacrophages.Sequelaewereassociatedwithearlydecreasesincompoundmuscle-actionpotentialamplitudes,indicatingimpairedfunctioningofeitherproximalsciaticnerveaxonsand/orneuromuscularsynapses.Spinalsomatosensory-evokedpotentiallatenciesalsowereincreased,indicatingimpairedsomatosensoryfunctionatthesciaticnerve,dorsalroots,spinalcord,and/orpostsynapticinterneurons,althoughthepreciselocationofimpairmentcouldnotbedelineated.Similaritiesbetweenthissyndromeandimmune-mediatedpolyneuropathiesinhumansarediscussed.
