Purpose: To investigate the biological effect of IL-17/NF- B immune inflammatory pathway on pathological damagein synovium tissue of Rheumatoid arthritis (RA) and study inregulation mechanism of Chinese herbal compound Tengmeidecotion for IL-17/NF- B signal pathway.Methods: To establish collagen II-induced rats arthritis(CIA)models. The successful models of SD rats wererandomly divided into model group, positive group, and high-,medium- groups of Chinese medicine, 6 rats in each group. Thenormal control and model groups were given distilled water(10ml kg-1 d) by gavage. The positive drug group was givenleflunomide (1.87g kg-1 d) by gavage. The high and mediumdose Chinese medicine groups were given crude medicine of31.8 g kg-1 d and 15.9 g kg-1 d by gavage. After twelve weeksof treatment intervention, all SD rats were executed, theblood and synovium tissue samples were kept for detectingIL-17, NF-KB P65 mRNA transcription and protein expressionby RT-PCR, WESTEN BLOT and ELISA Analysis, and detectinginflammatory infiltration in synovium tissue by Histopathologicalanalysis.Results: (1)Compared to the normal control group, levelsof mRNA transcription and protein expression of IL-17and NF- B P65 were significantly up-regulated (P < 0.01) in themodel group. Compared to the model group, levels of mRNAtranscription and protein expression of IL-17 and NF- B P65were significantly down-regulated in the positive and Chinesemedicine groups (P < 0.01).(2) Histopathological analysis displayedthat mild hyperplasia of epithelial cells covering thearticular cartilage synovium, joint cavity narrowing, and mildinflammatory lymphocytes infiltration in model group, withjoint lesions improved in treatment groups.Conclusion: The molecular mechanisms of Chinese herbalTengmei decotion in inhibiting immune inflammatory pathologicaldamage in synovium of CIA rats models related to itseffects on IL-17/NF- B pathway.
