Wedevelopedanovelapproach,J-bindingprotein1sequencing(JBP1-seq),thatcombinesthebenefitsofanimprovedrecombinantJBP1protein,Nextera-basedlibraryconstruction,andnext-generationsequencing(NGS)forgenome-wideprofilingof5-hydroxymethylcytosine(5hmC).ComparedwiththeoriginalJBP1,thisnewrecombinantJBP1wasbiotinylatedinvivoandconjugatedtomagneticbeadsviabiotinndash;streptavidininteractions.Thesemodificationsallowedamoreefficientandconsistentpull-downofβ-glucosyl-5-hydroxymethylcytosine(β-glu-5hmC),andsequence-readylibrariescanbegeneratedwithin4.5#xA0;hfromDNAinputsaslowas50#xA0;ng.5hmCenrichmentofhumanbrainDNAusingthenewJBP1resultedinover25,000peakscalled,whichissignificantlyhigherthanthe4003peaksenrichedusingtheoldJBP1.Comparisonofthetechnicalduplicatesandvalidationswithotherplatformsindicatedtheresultsarereproducibleandreliable.Thus,JBP1-seqprovidesafast,efficient,andcost-effectivemethodforaccurate5hmCgenome-wideprofiling./p/div
