GABAERGIC POTENTIATION OF REDUCED BOWELMOVEMENTS INDUCED WITH ZINC SULPHATE BY BACLOFEN

This study evaluated the influence of orally administered zinc sulphate on bowel movement of rats by the faecal outputand gastrointestinal transit time in the rats. The effects of zinc sulphate on feeding as well as water intake were alsodetermined. The dose of zinc sulphate that yielded the maximal effect was administered alongside baclofen toinvestigate the role of GABA on bowel propulsion. All the three doses of zinc sulphate (1 mgkg-1, 5 mgkg-1 and 10mgkg-1) produced significant reduction (p < 0.05) in total faecal pellets produced in eight hours of study (12.00 ± 0.93,9.17 ± 0.31 and 3.17 ± 0.17) respectively, compared with the control group (15.67 ± 1.38). Also, the three doses ofzinc sulphate produced dose-dependent significant reduction (p < 0.05) in the faecal mass (1.78 ± 0.17, 2.50 ± 0.22and 1.07 ± 0.10) respectively when compared with control group (3.39 ± 0.25).The total transit time of zinc sulphate(10 mgkg-1) treated group (593.8 ± 2.84 min) increased significantly (p < 0.0001, t = 26.98) compared with the controlgroup (251.2 ± 5.48 minutes). Zinc sulphate (5 mgkg-1 and 10 mgkg-1) significantly (p < 0.05) reduced food intake(10.83 ± 0.54 and 12.50 ± 0.51) respectively while (1 mgkg-1) produced increase (p < 0.05) in food intake (22.50 ± 0.56)when compared with the control group (18.33 ± 0.67). Administration of zinc sulphate and zinc + baclofen significantly(p < 0.001, f = 63.9) reduced faecal pellet output (4.80 ± 0.74 and 1.60 ± 0.25) respectively when compared withcontrol (10.2 ± 0.74). The study concluded that zinc sulphate reduced bowel movement in rats stimulating γaminobutyric acid (GABAB) receptors
