AntigenemergencerapidlystimulatesTcells,whichleadstochangesincytokineproduction,cellproliferation,anddifferentiation.Someofthekeymoleculesinvolvedintheseevents,suchasTGF‐β1andNOTCH1,aresynthesizedinitiallyasinactiveprecursorsandareproteolyticallyactivatedduringTcellactivation.PCSKsregulateproproteinmaturationbycatalyzingtheproteolyticcleavageoftheirsubstrates.TheprototypePCSKFURINisinduceduponTCRactivation,anditsexpressioninTcellsiscriticalforthemaintenanceofperipheralimmunetolerance.Inthisstudy,wetestedthehypothesisthatFURINregulatesTcellactivation.OurdatademonstratethatIL‐2isincreasedinitiallyinFURIN‐deficientmouseCD4+Tcells,buttheTCR‐inducedIL‐2mRNAexpressionisnotsustainedintheabsenceofFURIN.Accordingly,theinhibitionofFURINinhumanJurkatTcelllinesalsoresultsinadecreaseinIL‐2production,whereastheoverexpressionofWTFURINisassociatedwithelevatedIL‐2levels.InJurkatcells,FURINisdispensableforimmediateTCRsignalingsteps,suchasERK,ZAP70,orLATphosphorylation.However,withtheuseofgenereporterassays,wedemonstratethatFURINregulatestheAP‐1,NFAT,andNF‐κBtranscriptionfactors.Finally,byperformingatranscriptionfactor‐bindingsiteenrichmentanalysisonFURIN‐dependenttranscriptomes,weidentifytheFURIN‐regulatedtranscriptionfactorsinmouseCD4+Tcellsubsets.Collectively,ourworkconfirmsthehypothesisthattheTCR‐regulatedproteaseFURINplaysanimportantroleinTcellactivationandthatitcanspecificallymodulateTCR‐activatedtransactivation...
