MacrophageadherencetotheinnercornealsurfaceandformationofMGCsinthestromaarecommonsignsofchronicinflammationfollowingcornealinfection.Todeterminewhethermacrophageadherence(knownclinicallyasKPs)andgiantcellformationwerespecifictoinnateimmuneactivationviaparticularTLRligands,macrophageactivationwasexaminedinamurinemodelofTLR‐mediatedcornealinflammation.ThecornealepitheliumwasdebridedandhighlypurifiedTLRligandsweretopicallyappliedoncetothecorneaofTLR7?/?,TLR9?/?,Cx3cr1gfp/+,CD11ceYFP,andIL‐4?/?mice.At1weekpost‐treatmentmacrophageactivationandphenotypewasevaluatedinthecornea.TreatmentwithTLR2,TLR3,TLR4,andTLR5ligandscausedanincreaseinthenumberofactivatedstromalmacrophagesinthecentralcorneaat1weekpost‐treatment.However,treatmentwithTLR9ligandCpG‐ODNandtheTLR7/8ligandR848/Resiquimodledtoanaccumulationofmacrophagesonthecornealendotheliumandformationofmultinucleatedgiantmacrophagesinthecornealstroma.Wesuggestthatgiantcellformation,whichisacharacteristicfeatureofgranulomaformationinmanytissues,maybeauniquefeatureofTLR9‐andTLR7/8‐mediatedmacrophageactivation...
