Duringpregnancy,aspecializedtypeofNKcellaccumulatesintheliningoftheuterus(decidua)andinteractswithsemiallogeneicfetaltrophoblastcells.dNKcellsarefunctionallyandphenotypicallydistinctfromPBNKandareimplicatedinregulationoftrophoblasttransformationoftheuterinespiralarteries,whichifinadequatelyperformed,canresultinpregnancydisorders.Here,wehaveuseduterinearteryDopplerRIinthefirsttrimesterofpregnancyasaproxymeasureoftheextentoftransformationofthespiralarteriestoidentifypregnancieswithahighRI,indicativeofimpairedspiralarteryremodeling.WehaveusedflowcytometrytoexaminedNKcellsisolatedfromthesepregnanciescomparedwiththosefrompregnancieswithanormalRI.WereportareductionintheproportionofdNKcellsfromhighRIpregnanciesexpressingKIR2DL/S1,3,5andLILRB1,receptorsforHLA‐CandHLA‐Gontrophoblast.DecreasedLILRB1expressioninthedeciduawasexaminedbyreceptorblockingintrophoblastcocultureandaltereddNKexpressionofthecytokinesCXCL10andTNF‐α,whichregulatetrophoblastbehavior.TheseresultsindicatethatdNKcellsfromhighRIpregnanciesmaydisplayalteredinteractionswithtrophoblastviadecreasedexpressionofHLA‐bindingcell‐surfacereceptors,impactingonsuccessfultransformationoftheuterusforpregnancy...
