Thegapjunctionfamilyofproteinsiswidelyexpressedinmammaliancellsandformintercellularchannelsbetweenadjacentcells,aswellashemichannels,fortransportofmoleculesbetweenthecellandthesurroundingenvironment.Inaddition,gapjunctionproteinshaverecentlybeenimplicatedasimportantfortheregulationofcelladhesionandmigrationinavarietyofcelltypes.Thegapjunctionproteinconnexin43(Cx43)regulatesBlymphocyteadhesion,BCR‐andLFA‐1‐mediatedactivationoftheGTPaseRap1,andcytoskeletalrearrangementsresultinginchangestocellshapeandmembranespreading.WedemonstrateherethattheactincytoskeletonisimportantforthedistributionofCx43intheBcellplasmamembraneandforothercellprocessesinvolvingthecytoskeleton.UsingshRNAknockdownofCx43inBlymphomacellsweshowthatCx43isalsonecessaryforchemokine‐mediatedRap1activation,motility,CXCL12‐directedmigration,andmovementacrossanendothelialcellmonolayer.TheseresultsdemonstratethatinadditiontoitsroleinBcellspreading,Cx43isanimportantregulatorofB‐cellmotilityandmigration,processesessentialfornormalB‐celldevelopmentandimmuneresponses...
