Intercellularjunctionformationinpreimplantationmouseembryoswasinvestigatedwiththin-sectionandfreeze-fractureelectronmicroscopy.Atthefour-cellstage,regionsofclosemembraneappositionwithfocalpointsofmembranecontactandoccasionalunderlyingcytoplasmicdensitieswereobservedbetweenblastomeresofthin-sectionedembryos.Correspondingintramembranespecializationswerenot,however,observedinfreeze-fracturedembryos.Atthe8-to16-cellstage,smallgapandmaculaoccludensjunctionsandcomplexesofthesejunctionswereobservedatalllevelsbetweenblastomeresoffreeze-fracturedembryos.Asdevelopmentprogressedfromtheearlytomid8-to16-cellstage,thesizeoftheoccludens/gapjunctioncomplexesincreased,formingfasciaoccludens/gapjunctioncomplexes.Atthemorulastage,gapjunctionsandoccludens/gapjunctioncomplexeswereobservedonbothpresumptivetrophoblastandinnercell-masscells.Zonulaoccludensjunctionswerefirstobservedatthemorulastageonpresumptivetrophoblastcellsoffreeze-fracturedembryos.Thenumberofembryospossessingzonulaoccludensjunctionsincreasedatthemidcomparedtotheearlymorulastage.Attheblastocyststage,junctionalcomplexesconsistingofzonulaoccludens,maculaadherens,andgapjunctionswereobservedbetweentrophoblastcellsoffreeze-fracturedandthin-sectionedembryos.Isolatedgapandoccludensjunctions,adherensjunctions,andoccludens/gapjunctioncomplexeswereobservedontrophoblastandinnercell-masscells.
