Wehaveexaminedthemorphologyoffetalratsympatheticneuronsgrowninserum-freemediumintheabsenceofnonneuronalcells.Becausecelldensitycanaffectphenotypicexpressioninvitro,themorphologicalanalysiswassubdividedintothestudyofisolatedneurons(neuronswhosesomatawereatleast150??mfromtheirnearestneighbor)andofmorehighlyaggregatedneurons.Whenisolatedneuronswereinjectedwithintracellularmarkers,itwasfoundthatmost(79%)hadasingleprocessemanatingfromtheirsomataandthatthisunipolarstatepersistedforatleast8weeksinvitro.Theprocessesofunipolarsympatheticneuronshadtheappearanceofaxonsinthattheywerethinandlong,hadaconstantdiameter,andwererelativelyunbranched.Cytochemicalmethodsrevealedthatsuchprocesseshadotheraxonalcharacteristics:(1)theyweremorereactivewithamonoclonalantibodyagainstphosphorylatedformsoftheMandHneurofilamentsubunitsthanwithanantibodytononphosphorylatedformsoftheseproteins;(2)theyalsoreactedwithantibodiestothe??microtubule-associatedproteinandtothephosphorylatedformsoftheHneurofilamentsubunit;and(3)theycontainedonlysmallamountsofRNAasdeterminedby[3H]uridineautoradiography.Thesedataindicatethatneuronswhichnormallyformdendritesinvivoneednotexpressthiscapacityinvitroandthataxonalanddendriticgrowthcanbedissociatedundersomeconditionsinculture.Whilemostisolatedneuronswereunipolar,neuronsinregionsofhighneuronalcelldensitywereusuallymultipolar.Inadditiontoaxons,multipolarneuronshadprocesseswithsomeofthecharacteristicsexpectedofrudimentarydendrites:theyendedlocally(usuallywithin100??m),wereoftenhighlybranched,andreactedwithanantibodytononphosphorylatedformsoftheMandHneurofilamentsubunits.Theeffectsofdensityweremostprominentwhenneuronswerewithinaggregatesinwhichthesomatawereincloseapposition.Density-dependentchangesinmorphologywerelessfrequentlyobservedwhenneuronalsomatawereseparatedbygreaterdistances(30a€“100??m).Thesedataindicatethatthemorphologyofsympatheticneuronsissubjecttoenvironmentalregulationandthatneuron-neuroninteractionscanpromotetheextensionofrudimentarydendritesinvitro.
