Muralandcumulusgranulosacellssynthesizehyaluronicacid(HA)andexpandinvitroinresponsetofollicle-stimulatinghormoneandasolublefactor(s)producedbyfullygrownoocytes.InthepresentstudyweexaminedHAsynthesisandextracellularmatrixorganizationbythetwocellpopulationsinvivoduringthepreovulatoryperiod.Afterinjectionofhumanchorionicgonadotropinintopregnantmares'serumgonadotropin-primedanimals,aprogressiveincreaseinHAsynthesiswasobservedbythecumuluscell-oocytecomplex(COC),andbythemuralgranulosacellsadjacenttotheantrum(antralgranulosacells).Theoutermostlayersofmuralgranulosacells(peripheralgranulosacells)didnotsynthesizeHA.NetHAsynthesiswasa??4pg/cellforCOCsisolatedafterfullexpansioninducedeitherinvivoorinvitro,whereasthetotalHAcontentandcellnumberintheovulatedCOC(a??11ngHAanda??3000cellsperCOC)wereaboutthreefoldhigherthanforCOCsexpandedinvitro(a??4ngHAanda??1000cellsperCOC).TheincreasedcellcontentofovulatedCOCsappearstobeprimarilytheresultofinclusionofproximalmuralgranulosacellswhichsynthesizeHAinresponsetotheoocytefactor(s)andbecomeincorporatedintheexpandedCOCextracellularmatrixmass.Mediaconditionedbyoocytesenclosedinthecumuluscellmass(intactCOCs)containedonly10a€“20%oftheHA-stimulatoryactivityofmediaconditionedbyanequalnumberofisolatedoocyteswhentestedonmuralgranulosacellcultures.Further,HA-stimulatoryactivityofmediaconditionedbyisolatedoocyteswasdramaticallyreduced(a??70%)bypreincubationfor5hrwithcumuluscellscomparedtopreincubationintheabsenceofcells.TheresultssuggestthatdifferencesinHAsynthesisbetweensubregionsofmembranagranulosadependonadiffusiongradientoftheoocytefactor(s).
