Preantralgranulosacells(PAGCs)differentiateintocumuluscellsfollowingantrumformation.Cumuluscells,butnotPAGCs,arecompetenttoundergoexpansion.ExperimentsreportedheretestedtherespectiverolesofbothoocytesandFSHinthetransitionofpreantralgranulosacellstocumuluscellscompetenttoundergoexpansion.PAGCndash;oocytecomplexeswereculturedwithorwithoutalowdoseofFSH(0.005nbsp;IU/ml)andisolatedPAGCswereculturedwithorwithoutoocytes.Attheendofculture,complexesorisolatedPAGCsweretestedfortheirabilitytoundergocumulusexpansionandupregulateexpansiontranscriptsinresponsetoEGForFSH(0.5nbsp;IU/ml).TheabilitytoundergoexpansioninresponsetoEGFrequiredthepresenceofoocytesbutnotFSHduringthecultureperiod.Likewise,complexesisolatedfromtheovariesofhypogonadalmice,whichlackcirculatinggonadotropins,underwentexpansioninresponsetoEGF,butnotFSH.Incontrast,theabilitytoactivateMAPK3/1andMAPK14andundergoexpansioninresponsetoFSHrequiredpriorexposuretolowdosesofFSH.However,theselowlevels(0.005or0.025nbsp;IUFSH/ml)suppressedexpressionofemSlc38a3/emandemAmh/em,twotranscriptshighlyexpressedincumuluscells,suggestingopposingeffectsofFSHoncumuluscelldifferentiation.Inconclusion,theabilitytoundergoexpansioninresponsetoFSHrequirespriorexposuretoFSHduringdevelopment,whileoocyte-derivedfactorsalonearesufficienttopromotetheabilitytoundergoexpansioninresponsetoEGF.TheseresultshighlightthecrucialroleofoocytesindrivingthedifferentiationofPAGCsintocumuluscellsduringthepreantraltoantralfollicletransition./p/div
