The orbital sinus puncture under anesthesia or tail vein incision method have been widely used for multiple blood collection from mice. However, these methods have disadvantages on animal well-being for pain refinement and data reliability for physiological change by some anesthesias. We developed the alternative blood collection technique using butterfly needle mounted capillary from tail vein in unanesthetized mice. Male ICR mice were used in this experiment. Blood collections were performed 6 times in 2 hours from tail vein at 7, 8 and 9 weeks of age. Collecting volume was 75uL (fill up capillary) at each time. The required time for blood collecting, glucose and hematocrit(HCT) were measured. It takes 30~120 seconds for blood collecting in approximately 90% of occasion. Blood glucose level and HCT were comparable in each age. Congestion was observed at the tail on the next day of blood collection, but disappeared within 5 days. These results suggested that our method with this needle is available for multiple blood collection in mice with less physical damage.
