Improvement of medium for in vitro fertilization of inbred BALB/c mice

In vitro fertilization (IVF) of mice is used in reproduction research. However, the collection of embryos is often difficult because some strains show poor fertilization rates on IVF. The present study was performed to obtain IVF medium capable of supporting many strains. We compared the fertility rates in media consisting of modified human tubal fluid (mHTF) with and without addition of sodium lactate and containing various concentrations of calcium and glucose. The results of this study indicated that fertility improved with increased concentrations of calcium and glucose in the medium. In contrast, the fertilization rate decreased with the addition of sodium lactate during sperm capacitation. Based on these results, we examined sperm capacitated and inseminated medium, and have performed IVF with a stable high fertility rate of 70.3-91.5%. In addition, the IVF embryos showed ontogeny at a rate of more than 50%. Therefore, this medium appears to be effective for the preservation of strains and for production of the BALB/c strain.
