Genetic study on ocular coloboma in FLS mice

Coloboma refers to a defect of some of the ocular tissue (iris, retina, choroid, and/or optic nerve). Although ocular coloboma is detected in a small number of outbred albino mice, the incidence is almost 70% in FLS mice. Our previous studies clarified that the mice showed characteristic morphological features of coloboma in fundus during the postnatal period, and that a failure of basement membrane disintegration involved in failure of closure of the optic fissure in FLS mice. In the present study, the genetical analyses are undertaken on ocular coloboma of this strain. FLS mice with ocular coloboma were crossed with BALB/cAnNCrlCrlj or CBA/JNCrlj mice to create F_1 progeny. Then, F_1 females were backcrossed to FLS males to obtain N_2 mice. These N_2 mice were phenotyped for the ocular coloboma and genotyped with 104 microsatellite markers covering the genome to analyze linkage between ocular coloboma and marker loci.
