animal feeds that can impair rumen fermentation athigh concentrations; however, its effects at physiologicallyrelevant concentrations are unknown. This studyexamined the effects of aflatoxin B_1 (AFB_1), with orwithout bentonite clay (CL) and Saccharomyces cerevisiaefermentation product (SCFP)–based sequesteringagents on in vitro rumen fermentation and digestibilityof a dairy cow TMR. Corn silage-based TMR (0.5 g,17.3% crude protein and 1.67 Mcal/kg of net energy forlactation) was incubated in a rumen fluid-buffer inoculum(1:2 ratio; 50 mL) with the following treatments:(1) no additives (control); (2) control + 0.75 μg/LAFB_1 (T); (3) T + 80 mg/L sodium bentonite clay(CL; Astra-Ben-20, Prince Agri Products Inc., Quincy,IL); or (4) CL + 14 mg/L SCFP (CL+SCFP; DiamondV, Cedar Rapids, IA). Ruminal fluid was collected 3 hafter the morning feeding from 3 cannulated cows fedthe same TMR, and rumen fluid from individual cowswas used to prepare separate inocula. Each treatmentwas incubated in duplicate at 39°C for 0, 4, 8, 16, and24 h in each of 3 runs. Adding T reduced total volatilefatty acid (VFA) concentration after 4 and 8 h andmolar proportion of propionate after 4 and 24 h of incubationrelative to control. Adding sequestering agents(CL and CL+SCFP) with T did not affect total VFAconcentration after 4 or 8 h, but increased total VFAafter 16 h and tended to increase molar proportion ofpropionate after 24 h compared with T. At 24 h, T hadlower DM digestibility and higher NH_3-N concentrationcompared with the control. Thus, AFB_1, even at verylow concentration (0.75 μg/L), had detrimental effectson rumen fermentation and subsequently DM digestibilityof the TMR. Adding sequestering agents did notprevent negative effects of T on rumen fermentationwithin 8 h of incubation; however, sequestering agentswere effective after 16 h of incubation.
