Statins are widely prescribed for lowering plasma low-density lipoprotein concentrations, thereby reducing the risk of cardiovascular disease, but response to treatment varies and there is concern about potential adverse effects. Lara Mangravite et at. screened for the effects of in vitro statin exposure on genetic associations with gene expression levels in lymphoblastoid cell lines derived from participants of a clinical trial of simvastatin treatment. They identified six expression quantitative trait loci that interact with simvastatin exposure, including one that affects GATM, a gene encoding glycine amidinotransferase-a rate-limiting enzyme in creatine synthesis. They also provide evidence to suggest that GATM acts as a functional link between statin-mediated cholesterol reduction and susceptibility to myopathy, a common side effect of statin therapy.
