How do arms or wings (forelimbs) become different from legs (hindlimbs)? This biological problem has interesting evolutionary implications, and a handful of papers — including reports by Takeuchi et al. and Rodriguez-Esteban et al. on pages 810 and 814 of this issue — provide the first tantalizing molecular clues. They show that two transcription factors, Tbx5 and Tbx4, which are expressed in the developing fore-limb and hindlimb, respectively, control the identity of arms and legs.
