The Gram-positive bacterium Streptococcus pneumoniae (pneu-mococcus) poses a serious threat to immunocompromised populations. Vaccines that target bacterial cap-sular polysaccharides, a coating that masks the pathogen from the host's immune system, provide serotype-specific immunity; but with more than 90 known capsule serotypes, devising effective vaccination strategies has proven difficult. In an Inaugural Article, Richard Malley and Porter Anderson (pp. 3623-3627) describe experimental serotype-independent pneumococcal vaccines that activate toll-like receptors and induce both cellular and humoral immunity in mice. The authors report two contrasting approaches: killed cells of a noncapsulated strain, which induce the production of antibodies to multiple non-capsular antigens and promote a type of immunity that may develop naturally in infants;rnand a biochemically defined, synthetic antigen that results in similar bifunctional activity.
