The malarial parasite Plasmodium thrives in mammalian hosts by multiplying within circulating red blood cells. The mature, blood-infecting stage of the parasite life cycle, called merozoite, arises in host liver cells from another stage, called sporozoite, injected into the skin throughrnmosquito bites. Since 1948, when researchers demonstrated that sporozoites transform into merozoites in liver cells, it has been widely held that liver cells are the only site for parasite maturation in mammals. By using confocal microscopy, Pascale Gueirard et al. (pp.18640-18645) studied the fate of genetically engineered, fluorescent, rodent-infecting sporozoites introduced into the ear skin of mice by microinjection or through Anopheles mosquito bites.
