Creating Biosensors - an Innovative Avenue in Transgenic Fish Research

Pollutants originating largely from industrial waste and sewage often cause reproductive dysfunction in fishes. The biochemical responses of organisms to organic and metal compounds in water (including pesticides) can be measured and used as a biomarker for the level of pollution. By using a heavy metal inducible promoter, it has been proposed that a transgenic fish-based biosensor system may be developed for monitoring aquatic pollution and detecting low levels of chemical contaminants that cause oxidative stress in aquatic environments. Biosensors/bioreporters are simple first-level screening organisms that can express a reporter protein in response to toxic levels of a specific analyte. In a conceptual approach, the transgenic fish would be placed in water containing a chemical to be tested. Following uptake, distribution and accumulation of it in fish tissues, the genomically integrated responsive elements would be activated and the reporter gene would be upregulated. The fish will be assayed for reporter gene activity that would be comparative to concentration of the chemical to which the fish got exposed.
