Memantine is used in Alzheimer's disease despite its uncertain benefits. This NMDA glutamate receptor antagonist is closely related to amantadine. Its adverse effects mainly include neuropsychological disorders linked to its atropinic and dopaminergic effects (1). In response to our request, the European Medicines Agency (EMEA) granted Prescrire access to a review of data on heart failure linked to memantine (2). In clinical trials, heart failure was more frequent with memantine than with placebo (0.90% versus 0.53%).
