Junctional complexes, including tight junctions, actin-associated adherens junctions, and intermediate-filament-associated desmosomes, are major structural components in epithelial cells that are essential for the establishment of epithelial cell polarity. Various signaling molecules, involving tyrosine kinases and phosphatases, small GTPases, and components of the Wnt signaling pathway regulate the assembly and disassembly of junctional complexes during embryonic development and cell differentiation. The levels of regulation involve the coordinated interactions of junctional proteins with the cytoskeleton, the interplay between different junctional complexes, and the active participation of junctional proteins in signaling cascades. The interaction of beta-catenin with both components of adherens junctions and proteins of the Wnt/wingless signaling pathway is the best characterized mechanism to date that links cell adhesion to gene expression and cell fate determination.
