Although some have expressed surprise that the Vibrio cholerae pathogenicity island (VPI) encodes a bacteriophage, our findings should not be too unexpected. The first suggestion of the phage-like nature of the VPI was the finding by Kovach et al. that the distal end of the tcpacf gene cluster contained a phagelike integrase gene and is inserted at a site homologous to the CP4-57 prophage integration site in Escherichia coli. A putative attachment (att) site was identified at the end of this region. Last year, we reported that the tcp and acf genes were part of a larger, approx 40-kb region that was absent from non-pathogenic strains of V. cholerae but present in all pathogenic strains. We sequenced a novel region of this locus and showed that the 'left' side contained an att site that was very similar (13/20 bp) to the att site at the 'right' side. This 40-kb region possessed other characteristics of a pathogenicity island and, in keeping with a trend in pathogenicity island nomenclature, we named it the VPI (Ref. 3).
