The aim of this study was to investigate the genetic characteristics of lymphocystis disease virus (LCDV) isolates from diseased Schlegel's black rockfish Sebastes schlegelii. Viral DNA was extracted from the skin lesions of rockfish and the major capsid protein (MCP) gene, adenosine triphosphatase (ATPase) gene and RNA polymerase 2 (domain 6) (Rpb2-6) gene were located, sequenced and compared with homologous genes from other LCDVs. DNA sequencing was performed using an automated DNA sequencer. Sequence analyses of the full-length ORFs of the three genes revealed the presence of genomic variants among LCDV isolates. Although one of the MCP gene sequences from rockfish was highly homologous (99%), the others from the flounder showed low sequence identity (80-85%) with the sequence data from the rockfish. The ATPase gene and the Rpb2-6 gene sequences from rockfish also showed low sequence identity (70-84%) with those from flounders. The MCP protein sequence from rockfish showed relatively high sequence identity (87-98%) to other LCDV isolates. However, there was only 71-86% identity in the ATPase protein and 77-88% in the Rpb2-6 protein. The findings revealed that LDCVs from rockfish showed low identities with those from flounder. The sequences of the MCP, ATPase and Rpb2-6 genes between rockfish and flounder were identical and strain variants existed.
