Diamond-like carbon coated polymer-based targets in microscope slide format for MALDI mass spectrometry

Since the introduction of matrix-assisted laser desorption/ionization (MALDI) mass spectrometry 20 years ago,[1,2] much effort has been put into the development of sample supports or targets to meet the users’ requirements for higher sensitivity, better reproducibility, selective enrichment or on-target sample cleanup.[3] Krutchinsky et al. report the use of a compact disc (CD) as a MALDI target and found CDs to be a promising replacement for traditional MALDI targets.[4] More recently, a novel composite target consisting of a polymer backing (a digital versatile disc), molybdenum interlayer and a diamond-like carbon (DLC) finish was presented, and its applicability as a sample support for LDI mass spectrometry was shown.[5] Besides this application to detect small molecule analytes without the interference from MALDI matrices and their fragments as well as clusters, DLC is also a promising material for MALDI target surfaces. Because of the mechanical robustness, chemical inertness and biocompatibility of DLC,[6,7] these targets could also be used in those cases where analyte interaction (e.g. phosphopeptides) with metal surfaces or corrosive additives render classical stainless steel sample supports unusable. Additionally, the design of these novel targets and the techniques applied for their production would make it possible to use them as disposables, thereby eliminating the problems inherent in such cleaning procedures as used for standard metal targets like mechanical damage, residues of detergents, change of surface properties, carryover from previously prepared samples and the consumption of solvents and time.
