Anxiolytic and nootropic properties of successive chloroform and methanolic extracts of Mitragyna parvifolia leaves is reported here. The test groups were orally administered the successive chloroform and methanolic extracts at 200 mg/kg and 400 mg/kg doses. The anxiolytic activity was studied using the mice models of elevated plus maze and basal locomotor score. The nootropic activity was evaluated using the rat model of elevated plus maze. The results were compared using the reference drug, diazepam (1 mg/kg orally) for anxiolytic activity and piracetam (100 mg/kg, orally) for nootropic activity determination. The successive chloroform extract showed significant anxiolytic activity in a dose-dependent manner. The anxiolytic activity of successive methanolic extract was also found to be significant at dose level of 400 mg/kg. However, the successive methanolic extract administered orally at doses of 200 mg/kg and 400 mg/kg orally produced a significant (P<0.05) dose dependent enhancement in cognition in the test animals. A significant decrease in anxiety produced by both the test doses of the successive chloroform extract showed potential anxiolytic action. The depression in anxiety and enhancement in the cognition of test animals treated with successive methanolic extract proved the usefulness of this extract both as a potential anxiolytic and nootropic agent.
